If you have only a single drive, then you cannot set up a RAID array. It requires multiple hard drives.
But if you're asking about using a single hard drive on the RAID connectors of the motherboard, then it depends on the board. A lot of the newer ATA133 RAID connections allow you to use them in a non-RAID setup. The motherboard documentation should cover what is necessary to do this.
