Are your headphones connected to the rear I/O panel? Headphone jack on your computer case?
Can you test with a different set of headphones?

I've tested 2 sets of cans. They both work on my laptop. It's like the other people who posted said; it seems like it's working but there's no sound.

The front panel connection doesn't work either. =(

Which front panel audio connector is connected to the motherboard? HD_Audio? AC'97?
In my system, HD_Audio is the default Realtek setting and there is an icon in the Realtek Sound Manager to use AC'97 instead.
Do you have speakers hooked up to your system? If yes, do they work?

In a few cases, I had no sound at all with my system, but it was due to incorrect sound manager settings with my Z-5500 speakers.

The front panel is HD audio as well. I'm going to try flashing my BIOS and see if that helps anything.

There has to be some way to fix this other than spending $50 on a PCI express sound card. I sure as hell don't want to buy a USB sound card; I've only heard bad things.

a bios flash is not going to help
also make sure the onboard audio is set as the default device in the windows mixer

It is set as the default, I covered every amateur mistake that there is already.

If you guys read the thread you'll see how confusing this problem is. The onboard detects when a jack is plugged in, it plays audio normally and shows that it's working, nothing is muted, but there is simply no sound making it to the headphones and the mic isn't recording although everything is set up properly.

Any news on this? Just put my new PC together last night and I have this same problem on the Z68 Fatal1ty Pro Gen3. The annoying thing is the audio WAS working during my initial setup of Win7 and all of a sudden it has stopped. Everything appears to be working properly, all devices are installed and Windows is picking up an audio signal but just no sound is delivered either via Analog to my headphones or SPIF to my speakers. Seems like it's conflicting with the onboard HDMI or Nvidia HDMI or something, I've tried all the suggestions in this thread so far and nothing has helped. It's really confusing, i would appreciate any help!

    Cheers

    I've this issue, too.
    My mobo: Asrock Z68 Extreme4 Gen3
    Bios: 1.10
    OS: Windows7 x64
Case: I try to install PCI board (IDE controller ST-Lab A160), but my pc freeze while booting (B2 phase on mobo led). In next step I remove PCI card and get no sound in windows. In Audio I see not connected Speakers and headphones.
I try connect speakers jack to another plug but I've still no sound.
I see digital audio connected (S/PDIF), but I have not any S/PDIF connection.

    Help me, please.

    P.S.: I use onboard video.
